Love is just a game of the brain.
I was standing in the forest waiting for her. Why forest? Because we met there for the first time and planned to meet at the same place after three years. I was all alone standing there just waiting for the time to pass so that I can meet my love, my destiny. She is the only thing, I have ever wanted from the so called almighty. I don’t believe in god but some incident happened in my younger days that has forced my mind to believe in the supernatural power of the almighty. There are not 33 crores god as said in the Hindu religion of course but some supernatural power is there governing the entire universe or some astrophysicist may quarrel with me it is not god but all about the string theory and so on. Faith differs as perception differs and so as individuality. Coming back to the track oh yeah I was waiting for her. The clouds of my sky were getting darker and denser. The birds were going back to their nest. The light of the sun was getting dimmer and dimmer. Then I realised it was no more an afternoon but a full-fledged evening. My love for her was so intense that I forgot about the time but was just eagerly waiting to see the face I hadn’t seen for years, the face which makes me smile, the face which has kept me young till date. The evening was getting even darker and soon pin drop silence prevailed among the woods. There was a fox roaring from a faraway place, may be it was a signal that it has got its prey for the night and it was a call to its fellow mates so as they can join the feast. There were no one with me but those small insects and frogs singing like anything unaware of my pain and sorrow. The world is a cruel place, your pain is only yours but your happiness is for everyone. The evening was then engulfed by midnight and my digital watch reminded me that it was 12 am in the morning. How a boy can wait this much but it was not just a boy but a lover waiting eagerly to bring their love together after few years of break.  All of a sudden my phone ranged and I found the name of my sweet heart on it with the first selfie we had taken as the contact photo. I picked it up like a thirsty crow throwing stones in the pot of quarterly filled water so as to relieve the drought prevailing in its throat. Suddenly my beloved from the other side said “ we can’t meet any more, this is all over, don’t ask me any questions what, why and when, just remember I love you and what I did is for our good only, never ever try to contact me. Just promise me one thing don’t shed tears for the person who is not even eligible for your love, Beep beep beep…… I dialled that number, number of times but it didn’t exist as the Jio connection said. I felt as if the gods out there both the old and the new are cursing on me at the same time and I am all alone getting troubled in the forest along with the insects and the frogs dancing in my pain.

Staining Of Microorganisms

From class 8 ,we have been studying about gram positive and gram negative bacteria and the color they produced on staining but we have hardly tried to find out the underlying reasons for the same. Let me reveal it...
actually there are 2 types of staining-
1.simple staining (only one type of stain is used) and 2.differential staining (more than 2 types of stain are used)
In simple staining :
we know that the cell wall of bacteria has negative charge due to the presence of phospholipid .So if we use acidic dye (acidic dye has negative charge as for e.g NEGROSIN), we have to stain the slide not the bacteria. Similarly if we want to use basic dye(which is positive as for e.g METHYLENE BLUE) then we can stain the bacteria itself not the back ground.

GRAM STAINING:
This Stain is composed of  1)CRYSTAL VIOLET 2)GRAHMS IODINE 3)ALCOHOL 4)SAFRANINE
Here (1) is the primary stain,(2) is the MORDANT(which helps in staining by increasing the affinity) ,(3)is the decoloriser and (4) is the secondary stain or counter stain.all these chemicals are added one by one after the fixation process (when we take the sample microbe in a slide and dry it and heat in flame so to evaporate the extra water content of cell)
when we add chemical (1) and (2) there is a formation of CRYSTAL VIOLET GRAM IODINE COMPLEX .
Now in case of GRAMS NEGETIVE BACTERIA when we add decoloriser i.e chemical(3) there is removal of the above mentioned complex and in this when we add chemical (4) we get PINK COLOR which is actually the color of SAFRANINE.
Similarily in case of GRAM POSITIVE BACTERIA when we add chemical (3) there is no removal of the above mentioned complex and so we do not get the effect of chemical (4) but we get the colour of CRYSTAL VIOLET which is of BLUE color.
So GRAM POSITIVE BACTERIA  gives BLUE color and GRAM NEGATIVE BACTERIA gives PINK color.
Now all this occurs due to the difference in composition of the cellwall of those bacterias,In GRAM +VE it has a thick layer of  PEPTIDOGLYCAN and in case of GRAM -VE it has a very thin layer of PEPTIDOGLYCAN.These composition made all the difference.
e.g of GRAM +ve BACTERIA ARE STAPHYLOCOCCUS AND STREPTOCOCCUS
e.g ofGRAM -VE BACTERIA ARE CYANOBACTERIA AND SPIROCHETES.
